The paper presents a novel microfluidic-based platform designed for the rapid detection and assessment of biofilm-associated infections (BAI), which are difficult to diagnose and treat with standard antibiotics. This label-free and multivariate analysis (LF-MA) system comprises a Biofilm Enrichment Device (BED) and a Severity Detection Device (SDD), enabling real-time biofilm enrichment and viscosity-based severity detection. The platform demonstrated high recovery efficiencies for various bacterial strains and introduced a new biosensor indicator, the HMS indicator, to evaluate infection severity based on microbead distribution and biofilm presence. Validated with clinical isolates, this one-step approach significantly reduces processing time to under two hours, aiming to enhance patient-centric management of BAI by facilitating quicker and more accurate assessments.
